| Abstract: | The emergence of COVID-19 led to a shortage of reagents and tests due to the demand for diagnostics against this disease. On the other hand, along with vaccination, there is a need to follow up the immunity of the population. For this reason, an ELISA type diagnosis is sought to be implemented locally to detect antibodies against the Spike of this virus. This proposal fulfills the objective of Good Health and Well Being of the UN and will allow to be prepared for the need of booster doses and diagnosis of future variants. A region of the spike was selected and cloned into a vector suitable for mammalian cells. It was transfected into HEK293 cells. Two Western Blots were performed, testing an AntiFlag and a positive serum against the virus. The plasmid construct was successfully obtained and verified by sequencing. The protein was detected only by the virus-positive serum; however, its size was smaller than expected. For the design of the ELISA assay, the variables to be considered were established, and a base assay was proposed. The obtaining of the genetic construct and the design of the ELISA represent a great achievement for the implementation of this immunodiagnostic locally, which will lead Ecuador to the sanitary sovereignty the country needs |
| Description: | Con la aparición del COVID-19se evidenció una escasez de reactivos y pruebas por la demanda de diagnósticos contra esta enfermedad. Por otro lado, con la vacunación surgió la necesidad de un seguimiento de la inmunidad de la población. Por esta razón se busca implementar un diagnóstico tipo ELISA localmente que detecte los anticuerpos contra la espícula de este virus. Propuesta que cumple con el objetivo de Salud y Bienestar de la ONU, y permitirá estar preparado ante la necesidad de dosis de refuerzo y de diagnósticos de futuras variantes. Se escogió una región de la espícula, y se clonó en un vector adecuado para células de mamíferos. Se transfectó en células HEK293. SerealizarondosWestern Blots, probando un Anti-Flag y un suero positivo contra el virus.La construcción del plásmido fue obtenida de manera exitosa, y comprobada por secuenciación. La proteína fue detectada solo por el suero positivo contra el virus, sin embargo, su tamaño fue menor al que se esperaba. Para el diseño del ensayo tipo ELISA se establecieron las variables a tomar en cuenta, y se propuso un ensayo base. La obtención deconstrucción genética y el diseño del ELISA representan un gran avance para la implementación de este inmunodiagnóstico de manera local, el cual encaminará al Ecuador a la soberanía sanitaria que necesita. |
