| Abstract: | The green iguana, Iguana iguana, is one of the iconic reptiles of the city; however, many aspects of its ecology are unknown including the number of populations that inhabit the region, the number of individuals that conform each population and the phylogenetic diversity of their intestinal microbiota. Previous studies suggested that this reptile belongs to the group of Reptiles Associated with Salmonellosis (RAS), which implies that its intestinal tract may be the natural host of Salmonella, thus, green iguanas are considered as a possible transmission vector of zoonotic and enteropathogenic diseases especially to humans that maintain close contact with this reptile. In order to dismiss the presence of this pathogenic bacterium, it was necessary to characterize the microbiota that inhabits the feces of green iguanas using microbiological, biochemical and machine learning approaches. Stool samples of 6 iguanas were collected in sterile containers, the feces samples were subsequently seeded for 24 h at 37°C in solid (general and selective) and liquid culture media. Six bacterial strains were isolated, purified and tentatively identified by the following biochemical tests: IMVIC, TSI and LIA. To detect the presence of Salmonella, the stool samples were enriched in peptone water and subsequently in RPV, the samples were incubated for 48 h at 42°C, then the samples that exhibited an abundant growth in RPV were seeded on XLD agar for 24 h at 37°C. The presence of Salmonella was not detected in any of the analyzed samples. The cellulolytic capacity of the bacteria that inhabit the feces was qualitatively determined in nutrient agar supplemented with 1% of CMC, a promising strain was isolated to be grown in selective CRC medium. The quantitative determination of cellulases was carried out in triplicate using test tubes containing 5 mL of the liquid selective medium CRC, the tubes were incubated for 96 h at 37°C, and the measurement of the released cellulases was performed by spectrophotometry at 48, 72 and 96 h using the method of quantification of reducing sugars by DNS. It was determined that the cellulolytic capacity of the promising bacterium was 17% lower than the positive control. |
| Description: | La iguana verde, Iguana iguana, es uno de los reptiles icónicos de la urbe, sin embargo, se desconocen muchos aspectos de su ecología, del número de poblaciones que habitan en la región, del número de individuos que conforman cada población y de la diversidad filogenética de su microbiota intestinal. Estudios previos sugieren que este reptil pertenece al grupo de Reptiles Asociados a Salmonelosis (RAS), lo que implica que su tracto intestinal es posiblemente un hospedero natural de Salmonella, convirtiéndolo en un posible vector de transmisiones de enfermedades zoonóticas y entero patógenas a los humanos que mantienen contacto directo e indirecto con este reptil. Para descartar la presencia de esta bacteria patógena, fue necesario caracterizar la microbiota que habita en las heces fecales de las iguanas verdes utilizando métodos microbiológicos, bioquímicos y de machine learning. Seis muestras de heces fecales fueron colectadas en envases estériles, las heces fueron posteriormente sembradas por 24h a 37° C en medios de cultivo sólidos (generales y selectivos) y líquidos. Se aislaron 6 cepas bacterianas, las cuales fueron purificadas e identificadas mediante pruebas bioquímicas: IMVIC, TSI y LIA. Para detectar la presencia de Salmonella se enriquecieron las muestras de heces en agua peptonada y posteriormente en RPV, las muestras fueron incubadas por 48 h a 42° C, transcurrido este tiempo, las muestras que crecieron en RPV fueron sembradas en agar XLD por 24h a 37° C. En ninguna de las muestras analizadas se detectó la presencia de Salmonella. La capacidad celulolítica de las bacterias que habitan en las heces fecales fue determinada cualitativamente en agar nutritivo suplementado con 1% de CMC, una cepa promisoria fue aislada y sembrada en medio selectivo CRC. La determinación cuantitativa de celulasas fue realizada por triplicado en tubos de ensayo que contenían 5 mL del medio selectivo líquido CRC, los tubos fueron incubados por 96h a 37° C, la medición de las celulasas liberadas se la realizó por espectrofotometría a las 48, 72 y 96 h utilizando el método de cuantificación de azucares reductores por DNS. Se determinó que la capacidad celulolítica de la bacteria promisoria fue 17% menor que la del control positivo. |
