dc.descriptionLa pitahaya es un cultivo que se ha vuelto trascendental en varios países, entre ellos Ecuador. Una de sus mayores limitantes es la incidencia de plagas y enfermedades que afectan su rendimiento. Éstas son provocadas por agentes fitopatógenos, los cuales incluyen hongos y bacterias, que han sido reportados en otros países y llegan a inducir pérdidas económicas importantes. No obstante, en Ecuador hay escasos estudios sobre estos microorganismos y sus perjuicios en la pitahaya. Por ello, el presente proyecto plantea determinar sus características e identificarlos, mediante estudios morfológicos y moleculares para levantar líneas base sobre su biología. Se recolectaron muestras de tejidos afectados de pitahaya en diversas localidades del país, se aislaron e identificaron los patógenos presentes, de los cuales se estudió dos: Phytophthora sp. y Diaporthe sp. Se obtuvieron cultivos puros de 3 aislados para ambos, y se evaluó su comportamiento en 3 incubadoras de: 24ºC, 28ºC y 30ºC, bajo luz LED. Asimismo, se extrajo ADN de cada aislado para una posterior identificación molecular. Se determinó que a 30ºC se dio la mayor tasa de crecimiento en la mayoría de aislados, pero las temperaturas y el tipo de aislado no influyeron significativamente en el crecimiento de ambos patógenos. Hubo escasa variación en las características morfológicas entre los aislados evaluados, los cuales formaron estructuras reproductivas asexuales y sexuales, predominando la formación de zoosporas y conidios alfa. Con la información obtenida, se logran sentar bases para que próximos estudios determinen la patogenicidad de estos fitopatógenos.-
dc.description.abstractPitahaya cultivation has become highly significant in several countries, including Ecuador. A major constraint is the incidence of pests and diseases that affect yield. These are caused by phytopathogenic agents—including fungi and bacteria—that have been reported elsewhere and can lead to substantial economic losses. However, there is limited research in Ecuador regarding these microorganisms and their impact on the crop. Therefore, this project aimed to characterize and identify them using morphological and molecular studies to establish baseline data on their biology. Samples of affected pitahaya tissue were collected from various locations across the country; the pathogens present were isolated and identified, with a specific focus on two genera: *Phytophthora* sp. and *Diaporthe* sp. Pure cultures were obtained from three isolates of each, and their growth behavior was evaluated in incubators set at 24°C, 28°C, and 30°C under LED lighting. Additionally, DNA was extracted from each isolate for subsequent molecular identification. The results showed that the highest growth rate for most isolates occurred at 30°C; however, neither temperature nor isolate type significantly influenced the growth of either pathogen. There was little morphological variation among the evaluated isolates; all formed both asexual and sexual reproductive structures, with a predominance of zoospores and alpha-conidia. The information obtained lays the groundwork for future studies to determine the pathogenicity of these phytopathogens.-
